The working principle of an autoclave is based on the principle of moist heat sterilization. The autoclave generates steam inside a closed chamber, increasing the pressure, which in turn raises the boiling point of water, allowing the steam to reach higher temperatures than boiling water at normal atmospheric pressure.

Autoclave Sterilization Process

The autoclave sterilization process involves several critical steps:

  1. Loading: Items to be sterilized are placed inside the autoclave chamber. Proper arrangement is crucial to ensure effective steam penetration.
  2. Sealing: The chamber is sealed to create a closed environment.
  3. Heating: The autoclave heats water to produce steam, which fills the chamber.
  4. Pressurizing: Once the steam is generated, the pressure inside the chamber is increased. Autoclaves typically operate at pressures of about 15 to 30 psi (pounds per square inch).
  5. Sterilization: The steam, under high pressure, reaches temperatures of 121°C to 134°C (250°F to 273°F). The high temperature and pressure effectively kill all microorganisms on the items.
  6. Depressurizing: After the sterilization cycle is complete, the pressure is gradually released.
  7. Drying: Some autoclaves include a drying cycle to remove moisture from the sterilized items.
  8. Unloading: The sterilized items are carefully removed, ready for use.
